چکیده

In transgenic mice, recombinant DNAsmicroinjected into pronuclei of fertilized eggs are generally integrated into host chromosomes.1} The integration mechanism of microinjected DNA is not fully understood because of limited information concerning the structures of integrated foreign DNAs(transgenes) and their flanking chromosomalDNAs.Transgenes of a few transgenic mice and their flanking regions have been cloned and analyzed.2~6) However, these methods included time-consuming steps, such as construction of genomic libraries, cloning, and subcloning. We have reported that polymerase chain reaction (PCR)7) and inverted PCR (IPCR)8~10) techniques were

design and evaluation PCR primer for type 1 tyrosinemia patient from yasuj

Tyrosinemia is a rare autosomal recessive genetic disease caused by fumarylacetoacetate hydrolase deficiency. 40 different mutation have been recognized related to Tyrosinemia that could be found in all extend of the gene with higher frequency from exon 8 to 14. Because of the size of FAH gene it's impossible to Sequence whole length of the gene by one round of sequencing reaction. Aim of this ...

Internal and flanking sequence from AFLP fragments using ligation-mediated suppression PCR.

Amplification fragment-length polymorphism (AFLP) analysis has proven to be a powerful tool for developing a large number of reliable genetic markers across a wide variety of organisms. Often it is desirable to further characterize these markers by obtaining internal and flanking sequence information. Here, we present a systematic approach for obtaining such information from AFLP markers. AFLP ...

Direct sequencing of single primer PCR products: a rapid method to achieve short chromosomal walks.

One of the limitations of PCR to analyse a region of interest is that it requires knowledge of the flanking sequences in order to design amplification primers. Several adaptations of PCR have been described which allow some unknown sequences adjacent to regions of known sequence to be amplified. Primers directed to repetitive sequences, for instance Alu repeats can be used to pair with a primer...

Sequence Characterization in 3′-Flanking Region of Bovine TNF-α: Association with Milk Production Traits and Somatic Cell Score in Holstein Cattle of Iran

Background: Tumor necrosis factor- a (TNF-α) is a cytokine that was identified as a factor with a wide range of proinflammatory activities. The expression of bovine TNF-α in mammary tissue during pregnancy seems to have a role in development of the corresponding glands. Objective: Single nucleotide polymorphisms (SNPs) were defined in 3′-flanking region of...

Heterozygote and mutation detection by direct automated fluorescent DNA sequencing using a mutant Taq DNA polymerase.

We describe a method for direct cycle sequencing of PCR fragments amplified from genomic DNA or cDNA. DNA sequencing template is amplified using PCR and oligonucleotide primers flanking the region of interest. The amplified fragment is directly cycle sequenced using fluorescent sequencing primers, Sanger dideoxy sequencing chemistry and an enzyme mixture of a mutant Taq DNA polymerase and therm...
